腫瘤組織研磨采用多樣品組織研磨儀進行其實驗效果如何？

實驗?zāi)康模?/strong>腫瘤組織提取蛋白，因手工研磨費時費力，在研磨過程中會破壞樣品活性，因此選擇上海凈信組織研磨儀。

實驗材料和器具：

樣品：腫瘤組織

工具：上海凈信多樣品組織研磨儀（Tissuelyser-24）

耗材：離心管，研磨珠

實驗步驟：

1、準(zhǔn)備好離心管及不同大小的研磨珠；

2、取腫瘤組織、剪成小塊與3顆研磨鋼珠分別放入離心管內(nèi)，加入裂解液，將蓋子扣緊，因為樣品從冰箱內(nèi)取出，如果馬上研磨將無法破碎，因此需要靜置片刻；

3、將適配器放入冰箱冷凍幾分鐘，以便在研磨過程中保持樣品低溫狀態(tài)，保證樣品活性；

4、首先將適配器放入研磨儀中固定，然后在將離心管放置適配器內(nèi)。樣品盡量對稱放置，以保證研磨時的受力平衡，旋緊垂直定位螺旋，將安全鎖扣緊。頻率及時間設(shè)定好后等待程序結(jié)束后即可得到破碎漿液；

5、研磨完畢后請關(guān)閉電源，將安全鎖歸位；

6、完成后續(xù)蛋白提取步驟。

注意事項：

1、每支離心管樣品量一般不超過放心管體積的三分之二。

2、樣品盡量對稱放置，以保證研磨時的受力平衡。
